Failure to prepare these documents does not ensure a person?s wishes are clearly documented. Regardless of the size of the estate, preparing an estate plan can help prevent quarreling among family members who believe they know what?s best for an individual?s care and how the estate should be handled after death.

Property Transfers Based on Non-Will Provisions. Most people think their Will controls who will get what when they die. However, many assets, such as bank accounts, certificates of deposit, retirement plans, annuities, life insurance policies and real estate are often not controlled by Wills. In the case of jointly-owned assets, the surviving joint owner often becomes the sole owner of the assets. Retirement plans, annuities and life insurance proceeds transfer to named beneficiaries, not necessarily to the people named in a Will. Why does this matter? Assets can flow to the wrong people due to old, wrong and/or out-of-date designations, often with unintended estate and income tax implications.

Online or Do-It-Yourself Rather Than Professionals. There has been a noticeable rise in the number of people who will look to the Internet to prepare their own Wills and Trusts. There are dozens of websites that will profess to offer you just the right discounted estate planning documents. Unfortunately, relying on web-based, do it yourself solutions is a recipe for disaster. Estate planning documents should represent the culmination of a well thought out financial and estate plan. Internet sites can provide you with documents, but no actual advice that fits in the context of your specific financial and personal life.

Leaving Assets Outright to Adult Children. There has been a growing opinion for families that assets should remain in trust, even for adult children, for as long as possible for asset protection and other benefits that a trust can offer. The bigger questions become who should really benefit from the fruits of my labor and how do I protect those assets from creditors, potential creditors and ex-spouses? Whether or not to leave assets in trust for adult children depends on many factors; not the least of which is personal preference.

You Have the Wrong Guardian Listed for Your Children. A Will is a mechanism to give instructions as to who you choose as guardians to care for your children. If you do not have a Will, your state decides who will care for them at a hearing. Check to see if your original guardian is still valid. A judge is required to act in the best interest of the child, so also consider writing a letter of explanation as to why you chose this guardian.

One of my favorite movies of the past year is a documentary by a new young Swedish filmmaker about a little-known (at least here) American musician of the '70s. It's called Searching for Sugar Man -- and it's a rock 'n' roll chronicle of a great singer/guitarist/songwriter about whom you've probably never heard named Rodriguez.

Back in the early '70s, he recorded two albums of his own songs -- 1970's Cold Fact and 1971's Coming From Reality, both of which had memorable music and strong guitar-playing and sharp, deeply poetic, socially hip lyrics (in the Bob Dylan mode) and both of which received very favorable reviews from critics. But they both flopped commercially, Rodriguez lost his recording contract (with Sussex), and his career seemed to vanish, along with Rodriguez himself.

Except...

Except in South Africa, where bootlegs of Rodriguez's albums began to circulate among the country's youthful music-lovers, where the records were eventually released by local entrepreneurs (who didn't know where Rodriguez was either), and where the socially conscious balladeer became a superstar as big as Elvis or the Beatles, and as admired as them, or Dylan or Springsteen -- and, for the youth of the country, especially the white youth, also a pop cultural symbol of resistance to racism and apartheid.

Rodriguez's song "I Wonder" became an anthem of the anti-apartheid generation, much as Dylan's "Blowin' in the Wind" had been an American anthem of the anti-war movement. In South Africa, Cold Fact was one of the three most popular of the era. (The others were Abbey Road and Bridge Over Troubled Water.) Without a huge hype machine, or a publicity apparatus of any kind, with only a few pictures of the star himself circulating (accenting his charismatic Latino looks, long black hair, dark glasses and air of gravity), he nevertheless conquered his audience, at least in South Africa. And, in one of the most curious twists of a tale full of them, he never knew of these triumphs while they were happening.

In the void of actual information, unsubstantiated stories began to circulate about the singer and his mysterious disappearance, including urban legends that he had committed suicide on stage after his last concert, by gunshot or by setting himself on fire. But what was the truth, the cold facts? Rodriguez's loyal fans and aficionados began increasingly to wonder, including the South African critic/music man who wrote the notes for the 1990's release of Coming from Reality, Stephen "Sugar Man" Segerman (his nickname comes from Rodriguez's song "Sugar Man"), who called in the notes for information about the pop idol's death or fate.

Another writer, Craig Bartholomew-Strydom, began to scour the Internet, looking for clues and witnesses. Finally he found out what happened, and where Rodriquez had gone. And Strydom and later filmmaker Malik Bendjelloul found out what kind of person had written and sung and played those songs -- and never apparently profited from them, while they were helping change a country half a world away.

Searching for Sugar Man is partly a fascinating mystery-detective story, and it's far more enjoyable if some mystery remains.

To Rome With Love (A-)
U.S.: Woody Allen, 2012, Sony Pictures Home Entertainment

Woody Allen puts himself back on the screen in To Rome With Love -- playing an old fool -- and I think the part has possibilities. Allen's character, which he plays to addled perfection, is Jerry, a retired, right-wing malcontent opera director-producer married to Phyllis (Judy Davis) and traveling to Rome for the wedding of their daughter Hayley (Alison Pill). Jerry, kvetching away, discovers that Hayley's fianc?, Michelangelo (Flavio Parenti), is very Euro-left-wing and that his daughter's prospective father-in-law, undertaker Giancarlo, is a brilliant amateur opera singer (played by the brilliant professional opera singer Fabio Armiliato) and that he sings brilliantly, but only when he's in the shower. So?

Coming right after Midnight in Paris, the biggest commercial hit of Allen's career -- and an unusual (for him these days) critical hit as well -- To Rome With Love tries to catch some of the same magic, milk the same Euro-delight. For me, it does. Like Midnight in Paris, it's a combination travelogue and comic romantic fantasy satire -- except that, in this case, Allen has concocted four funny episodes, instead of concentrating on just one.

It's somewhat like the Italian all-star anthology films of the '50s and '60s, omnibus movies like Yesterday Today and Tomorrow (with three segments by director Vittorio De Sica and screenwriter Cesare Zavattini, all starring Sophia Loren and Marcello Mastroianni ) and Boccaccio '70 (four episodes directed by De Sica, Fellini, Visconti and Mario Monicelli, all with different casts).

But instead of showing the separate episodes one after the other, Allen has interwoven them like a Robert Altman ensemble piece.

In one of the funniest episodes, Alec Baldwin plays (very amusingly) a famous, brilliant architect named John, who runs across a young talented but unknown architect named Jack (Jesse Eisenberg), who is a big fan of John's and who may also be his younger self. Just like Jerry Lacy's "Spirit of Humphrey Bogart" in Play It Again, Sam, John, who seems invisible to everyone but Jack, keeps hanging around offering sardonic and rude advice about how to conduct his love life -- as Jack shuttles irresponsibly around Rome between nice girlfriend Sally (Greta Gerwig) and Sally's even more irresponsible love-me-I'm-neurotic best friend, actress Monica (Ellen Page).

In the third episode, a little nobody named Leopoldo (played by Roberto Benigni), suddenly becomes a big somebody: a media darling and a paparazzi magnet, pursued and harassed and photographed and interviewed everywhere, for no special reason beyond the fact that the media world has decided to treat him like a celebrity. (He's "famous for being famous," according to one character.) This brief fable shows us Allen at his most profound.

In the last episode, the most Italian-vignettish of them all, Penelope Cruz plays drop-dead gorgeous lady-of-the-evening Anna, here working in the afternoon: a superbabe in a skintight red minidress who somehow gets her dance card mixed up and winds up in the posh Roman apartment of a crushingly na?ve honeymooner from the provinces, Antonio (Alessandro Tiberi) -- at the same time Antonio's sweet beautiful little wife Milly (Alessandra Mostranardi) gets lost in Rome, and winds up in the arms of hot-blooded movie matinee idol Luca Salta (Antonio Albanese, seething), as well as in part of the plot of Fellini's classic The White Sheik.

Meanwhile, when his relatives show up, none of whom have seen Milly, Antonio tries to pretend that Anna is his wife, and she tries to pretend that she's not everybody's favorite fantasy hooker -- despite the fact that a lot of the bigwigs they bump into include a lot of regulars on her prime client list. And there's also a randy hotel burglar running around. This is what they call commedia dell 'arte, or French bedroom farce, or maybe commedia all'italia. But all I can say is: It made me laugh.

The four episodes share a common theme. They're about the perilous consequence of wish fulfillment, or of answered prayers, spoken or unspoken. The four mini-movies are all done in Allen's usual dryly classical, deadpan elegant long-take style, shot by Darius Khondji, filled with crisp, witty, on-the-nose and super-bright dialogue. And the cast is the tops, especially Baldwin, Cruz, Armiliato, Benigni -- and Allen.

Manhunt under way for Phoenix gunman

At least three people were shot at an office complex in Phoenix on Wednesday, police there say, and a manhunt for the shooter is under way.

The building was evacuated and the suspect fled the scene, Sgt. Tommy Thompson said. The shooting--which occurred in the building's lobby--does not appear to be random act, Thompson said, though he would not say if the suspect knew the victims. Witnesses say they heard as many as 10 gunshots fired.

Three victims were transferred to John C. Lincoln Hospital with gunshot wounds, he said. Their conditions were not immediately known, but according to Phoenix's NBC affiliate, one was in "extremely critical condition."

At least three other people were transported to the hospital with minor injuries related to the shooting.

A SWAT team began canvasing a neighborhood in search of the suspected shooter described by Thompson as an "older" white male, possibly in his 60s.

?At this point we don?t believe he is in the area,? Thompson said. ?This doesn?t appear to be a random type of incident.?

The shooting occurred on the same day former Arizona Rep. Gabrielle Giffords--who was shot in the head during a mass shooting in Tuscon in 2011--testified at a Senate hearing on gun violence.

"Speaking is difficult but I need to say something important," Giffords said at the top of the hearing. "Violence is a big problem. Too many children are dying. Too many children. We must do something. It will be hard, but the time is now. You must act. Be bold, be courageous, Americans are counting on you."

Giffords' husband Mark Kelly, who also testified at the hearing, broke the news of the workplace shooting to lawmakers on Capitol Hill.

"While we are having this hearing, in Phoenix, Arizona, there has been what appears to be a shooting with multiple victims," Kelly said. "I certainly agree that universal background checks will go a long way to saving people's lives."

Jane Austen tourism booms as 'Pride and Prejudice' turns 200

When a classic love story has a big birthday, it?s time to have a ball ? literally.

Monday marks the 200th anniversary of the publication of ?Pride and Prejudice,? the Jane Austen novel that introduced feisty heroine Elizabeth Bennet, seemingly aloof Mr. Darcy and a world of manor homes and grand estates that turned England into a dream destination for readers around the globe.

Balls, dances and other special events are taking place this year to celebrate the bicentennial as Austen continues to lure travelers.

?She?s hugely important,? said Meredith Pearson, a spokeswoman for VisitBritain. The country?s national tourism agency maintains a guide for fans eager to experience the "Pride and Prejudice" lifestyle.

?We found that, to this day, because of the popularity of those stories, it?s something that visitors really, really enjoy doing,? Pearson said.

Even if you haven?t read the book, chances are you?re familiar with the plot of ?Pride and Prejudice? after catching the recent film version starring Keira Knightley, seeing ?Bridget Jones? Diary????a modern reboot of the novel ? or watching the blockbuster 1995 BBC mini-series starring Colin Firth, which led to a huge surge in visitors, Pearson noted.

No wonder, then, tourism is booming along Austen?s trail in England, a path that traditionally takes visitors to the resort city of Bath, the verdant landscapes of Derbyshire and the village of Chawton.

The author lived in Bath from 1801 to 1806 and chose the city as the setting for two of her novels. Today, about 60,000 people a year flock to The Jane Austen Centre, which chronicles her Bath experience, said Jennie Rogers, general manager of the attraction.

Besides taking in the Centre?s permanent exhibition and sampling Georgian biscuits, visitors can also try on fashions from Austen?s era ? empire-waist dresses, bonnets, gloves and parasols for the ladies; waistcoats, coats and top hats for the gentlemen.

?Our visitors become overwhelmed when trying on our replica costumes,? Rogers said. ?(They) love the romance of Jane's novels and the way in which they can relate to her character's dilemmas and issues???even today.?

To celebrate the 200th anniversary of ?Pride and Prejudice,? the Centre is hosting an ?International Readathon? on Monday, when dozens of actors, authors and Austen experts will read the book aloud in its entirety during a 12-hour period.

The city of Bath has put together a suggested itinerary for Austen fans, including taking a walk along the Royal Crescent, where the author often strolled, and enjoying afternoon tea in The Pump Room, which was a popular gathering spot during Austen?s era.

If you?d like to dress up and party Austen-style, there?s the Jane Austen Festival Regency Costumed Summer Ball on June 22, which features Regency dancing???the intricate choreography that requires ladies and gentlemen to form lines and circles, and twirl around and bow to each other.

?We hope to dance Mr. Beveridge's Maggot???a dance made popular by the BBC 1995 version of ?Pride and Prejudice,?? said organizer Jackie Herring.

The novel is also the theme of this year?s Bath Jane Austen Festival, scheduled to take place Sept. 13-21, complete with a Mr. Darcy lookalike contest, a masked ball and workshops on how to perfect the Regency look.

Fans who want to stroll through grand estates and gardens featured in the novels should head to the Derbyshire region of England where many of the Austen movies and mini-series were filmed. The district maintains a list of all the locations used in the productions.

Pearson thinks U.S. visitors love the sites because they?re so uniquely British.

?It?s part of that fairly tale, in a way. Because (the U.S. is) such a young country comparatively, the fact that there are these gorgeous old manor houses and country houses ? it really takes people away and it really catches Americans? imagination,? Pearson said.

One such home is Chatsworth House, said to have inspired Pemberley, Mr. Darcy?s estate in ?Pride and Prejudice.?

To see where Austen created some of her characters, visit her former home in Chawton, now transformed into Jane Austen?s House Museum.

This is where the author spent the last eight years of her life and wrote ?Mansfield Park,? ?Emma,? and ?Persuasion.? To celebrate the 200th anniversary of ?Pride and Prejudice,? the museum is displaying first editions of the book, original illustrations and the table where Austen worked.

British tourism officials?? who are still giddy about the Queen?s Diamond Jubilee and all the other events that drew visitors to the region last year???are hoping Austen?s legacy will continue the momentum in 2013.

'Zero Dark Thirty' Debate: Torture Has No Place in a Civilized Society

As I sat through the movie "Zero Dark Thirty," I shut my eyes in some of the "interrogation" scenes and reflected on the debate surrounding the movie. Some have said the movie shows torture did help in finding Osama bin Laden. Others have said the film reveals that torture really wasn't instrumental in tracking him down. To quote Hillary Clinton (on another topic), does it really matter???

Torture has no place in a civilized society. No matter the success or means to achieve a good end, torture (or as some leaders referred to it, enhanced interrogation techniques) reflects our darkest impulses and pulls all of us in this democracy to a dark place.

Would we as a society tolerate this kind of torture of our own citizens? I think not. Do we really deep down find that this behavior makes us a better people? I sure hope not. As I watched other folks in the theater in downtown Austin turn away at the water boarding and other torture techniques, I thought to myself if we have to turn away watching a movie then doesn't that really tell us that this is wrong, no matter all the rationalization about it helping to save lives and track terrorists.

I also thought about my son who served in the army for five years who speaks Arabic and actually looks a good bit like the main terrorist character (when he hasn't shaved for a bit) who was tortured through most of the first half of the movie. It turned my stomach to think of my son tied up like that or treated like that by some tribal group in Iraq (where he was deployed for nearly a year and a half) who might think he was the aggressor in their country.

Putting my son in that terrorist's place just for a moment actually opened a truth for me. If we believe as underlined in the movie "Lincoln" and in President Obama's Inaugural Address, that it is a self-evident truth that "all men are created equal," then how can we as a free compassionate "Christian" nation allow this to go on. Does "all men" just stop at our shores? Does it stop when some person or group does something horrendous to us? It is not in the niceties that our values are tested, but when something horrible and awful and undeserved happens that we truly know who we are.

The debate surrounding "Zero Dark Thirty" shouldn't be about whether the means of torture actually helped in achieving the good end of tracking down bin Laden. It should really be about whether any good ends justify use of means we would never accept or tolerate on our citizens. In the end, it is the means with which we live life or the means in which our society and leaders act, that will determine what we will end up like in this great journey of life.

I am not arguing that we don't have a right to defend ourselves and fight back when innocents are threatened. I come from a large Irish Catholic family in Detroit who deeply believes in protecting each other and the vulnerable in our society. In fact, I have one brother who retired from the Coast Guard and one who served in the Marines, as well as my son who enlisted on his own as a 19-year-old.

I am just saying that if for a moment you closed your eyes in the awful torture scenes and put your son or daughter in the place of that terrorist or a brother or sister or a niece or nephew, would you think those were acceptable techniques to extract information. I believe we as a people are better than that, and if we really believe that "whatever you do to the least of my brethren, you do unto Me," then we deep down know the answer. Civil liberty and decency and justice are meant for all men and women. And each person in the world is our sister and brother or son and daughter.

Stretching your body?s muscles will actually enhance the healing process.

Next to breathing and breath work, stretching is the most beneficial exercises we can do for better health.

Done in tandem, breath work and stretching are the best support for easing aches, pain, tension, headaches, tight muscles and unexplained pain elsewhere in the body.

The different types of stretching

Just as there are different types of flexibility, there are also different types of stretching. Stretches are either dynamic (meaning they involve motion) or static (meaning they involve no motion).

Dynamic stretches affect dynamic flexibility and static stretches affect static flexibility (and dynamic flexibility to some degree).

When done properly, stretching can do more than just increase flexibility.

Benefits of stretching include:

  • ?enhanced physical fitness
  • ?enhanced ability to learn and perform skilled movements
  • ?increased mental and physical relaxation
  • ?enhanced development of body awareness
  • ?reduced risk of injury to joints, muscles, and tendons
  • ?reduced muscular soreness
  • ?reduced muscular tension
  • ?increased suppleness due to stimulation of the production of chemicals which, lubricate connective tissues
  • ?reduced severity of painful menstruation (dysmenorrhea) in females

Things to remember when beginning a stretching routine:

  • Wear comfortable clothes that won?t restrict body movement
  • Stretching should be pain free; don?t force the body into difficult positions
  • Move into each stretch slowly and avoid bouncing, which can actually tear muscles
  • Stretch on a clean, flat surface which is large enough to move freely
  • Hold stretches long enough (20-30 seconds) to allow muscles or joints to become loose
  • Repeat the stretch, generally 5 ? 10 times

The most effective stretches

One good over all stretch for your body is to lie on the floor and gently bring your knees up to your chest. Once there, put a little pressure on your knees with two hands.

Gently raise your head up toward the knees. Breath, in, out, in out?..slowly. Stretch, then relax. Repeat.

The effect of stretching on healing

Long, slow static stretching will help to increase blood flow and circulation, eliminating stasis and help to warm and invigorate the muscles and in turn lubricate the ligaments and joints.

Hackers take over sentencing commission website

WASHINGTON (AP) ? The hacker-activist group Anonymous says it hijacked the website of the U.S. Sentencing Commission to avenge the death of Aaron Swartz, an Internet activist who committed suicide.

The website of the commission, an independent agency of the judicial branch, was taken over early Saturday and replaced with a message warning that when Swartz killed himself two weeks ago "a line was crossed."

The hackers say they've infiltrated several government computer systems and copied secret information that they now threaten to make public.

Family and friends of Swartz, who helped create Reddit and RSS, say he killed himself after he was hounded by federal prosecutors. Officials say he helped post millions of court documents for free online and that he illegally downloaded millions of academic articles from an online clearinghouse.

World's most powerful engine blazes path for space launch system advanced propulsion

Jan. 26, 2013 ? To help develop the nation's future heavy lift rocket, NASA resurrected the world's most powerful rocket engine ever flown -- the mighty F-1 that powered the Saturn V rocket-- and test fired its gas generator today at the Marshall Space Flight Center in Huntsville, Ala.

NASA engineers ran the gas generator at the Marshall Center's Test Stand 116. The test is part of a series that will push the gas generator to limits beyond prior Apollo-era tests. Modern instruments on the test stand measured performance and combustion properties to allow engineers a starting point for creating a new, more affordable, advanced propulsion system.

"Our young engineers are getting their hands dirty by working with one of NASA's most famous engines," said Tom Williams, Director of the Propulsion Systems Department in Marshall Engineering Directorate. "These tests are only the beginning. As SLS research activities progress, these young NASA engineers will continue work with our industry partners to test and evaluate the benefits of using a powerful propulsion system fueled by liquid oxygen and rocket grade kerosene, a propellant we haven't tested with in some time."

The gas generator tested at Marshall today is a key F-1 rocket component that burns liquid oxygen and kerosene and is the part of the engine responsible for supplying power to drive the giant turbopump. The gas generator is often one of the first pieces designed on a new engine because it is a key part for determining the engine's size, which is a factor in the engine's power and ability to lift heavy payloads and send them to space.

NASA's Space Launch System will provide an entirely new capability for human exploration beyond low Earth orbit. The initial 77-ton (70-metric-ton) SLS configuration will use two 5-segment solid rocket boosters similar to the boosters that helped power the space shuttle to orbit. The evolved 143-ton (130-metric-ton) SLS vehicle will require an advanced booster with more thrust than any existing U.S. liquid- or solid-fueled boosters. Last year, NASA awarded three contracts aimed at improving the affordability, reliability and performance of the rocket's advanced booster, including one focused on the F-1 engine.

"It's important that our workforce get hands on experience on systems like the F-1 gas generator as it helps make them smart buyers, and good stewards of what we procure from industry," said Chris Crumbly, manager of the SLS Advanced Development Office at the Marshall Center. "As we look to the future advanced boosters for SLS we are eager to see what our partners in industry can provide as far as a more powerful and affordable solution."

Now, we can certainly appreciate the beauty in the classical aesthetic that so often characterizes sites of religious observance. But sometimes even traditional venues need a radical makeover.

Graffiti artist Hense did just that to a former church in Washington D.C.'s up-and-coming arts district. The artist got to work with the help of a small crew, using rollers, brushes, spray paint, inks, acrylics, mops, enamels and paint sprayers to cover every inch of the edifice in popping hues. After several weeks, the white church was transformed into a rainbow splattered objet d'art.

Research may lead to new strategies against sepsis

Jan. 25, 2013 ? Scientists at the Center for Translational Medicine at the Temple University School of Medicine are inching closer to solving a long-standing mystery in sepsis, a complex and often life-threatening condition that affects more than 400,000 people in the U.S. every year. By blocking the activity of a protein, STIM1, in cells that line the insides of blood vessels in mice, they have halted a cascade of cellular events that culminates in the out-of-control inflammation that marks sepsis, and protected lungs from severe damage.

The findings, reported online January 25, 2013 in the Journal of Clinical Investigation, provide new insights into molecular details underlying sepsis and its effects on the lungs. In identifying STIM1 as a potential drug target, the results may lead to new treatment strategies against sepsis.

"While antibiotics are improving, more than 25 percent of those who develop sepsis will die from it. This is partly because we don't fully understand the mechanisms behind the widespread inflammation it causes," said senior author Muniswamy Madesh, PhD, Assistant Professor of Biochemistry at Temple University School of Medicine and a member of Temple's Center for Translational Medicine. "We've provided evidence indicating that without STIM1 driving calcium signaling, the exacerbation of inflammation can't occur. Our results could lead to a whole range of new therapeutic research directions."

STIM1 plays a variety of roles in the cells, including serving as a sensor for the amount of calcium inside a cell, and driving calcium signaling, which is important for cellular communication. In sepsis, a bacterial infection produces toxins that cause the body's normal reaction to infection to go haywire, prompting the immune system to attack the body's own organs and tissues.

According to Dr. Madesh, these toxins -- small molecules called lipopolysaccharides -- set off a chain of events. They initially bind to endothelial cell receptors, sending chemical signals that can result in "oxidative" damage to cells. STIM proteins can detect this cell damage, and drive the flow of calcium ions into the cell, increasing calcium signaling. The resulting fluctuations in calcium levels activate endothelial cells.

"When the endothelial cells are activated, they express various pro-inflammatory molecules, which facilitate white blood cells to adhere to endothelial cells and migrate from the blood to lung tissue," Dr. Madesh explained. "This cell migration further stimulates the immune system, increasing the release of other signaling molecules and factors. But how this occurs hasn't been completely understood."

Dr. Madesh and his colleagues wanted to better understand how STIM proteins were involved in lung inflammation and injury, which is commonly seen in sepsis. Such injury can lead to edema, or fluid, in the lungs, and possibly death. Their previous research had indicated that STIM1 played an important role in oxidative cell damage-altered calcium levels.

To find out, the researchers created mice lacking STIM1 in endothelial cells, and in a series of experiments, compared these mice to normal mice exposed to the sepsis toxin. They found that without STIM1 in the cells, the calcium fluctuations did not occur, and endothelial cells were protected against the toxin-induced lung injury.

The investigators also used a small molecule, BTP2, to see the effect on lung damage of physically blocking the STIM1 signaling pathway. "We found that the small molecule inhibitor blocked this calcium entry in the channel, as opposed to the other strategy in which the STIM1 gene was knocked out. Eliminating STIM1 or blocking the channel both reduced the permeability of the lungs' blood vessels and lessened lung edema. We can block this pathway using both genetic and pharmacological approaches, and both protected against endotoxin-induced lung inflammation," Dr. Madesh said.

"Although this STIM-mediated signaling pathway is essential for development and other functions, in the case of vascular inflammation, blocking the pathway protected that animal from the damage involved. While several other pathways have been established in the sepsis model, our finding is a new signaling pathway that could be targeted for therapeutic interventions."

Because BTP2 targets a specific calcium channel rather than the protein itself, Dr. Madesh said that one of his team's next goals is to "design new molecules that target the activation of the STIM protein." This same strategy could be used for other diseases or conditions, including stroke, that involve STIM-controlled calcium signaling, he noted.

Google&#39;s Kansas City experiment begins to yield start-ups | TribLIVE

KANSAS CITY, Kan. ? Smack in the middle of the nation, this city is about as far as possible from the hubs of high-tech innovation on both coasts.

An effort last spring to excite new Web entrepreneurs in a place better known for cattle drives and barbecue sauce turned up just a dozen people.

Then Google blew into town.

The company, dominant in the virtual world, began digging actual holes in the ground and connected homes and businesses to Internet speeds 100 times faster than most Americans have ever seen.

Three months into Google?s much-publicized experiment, signs of new business life have emerged. Nick Budidharma, an 18-year-old game developer, drove with his parents from Hilton Head, S.C., to live in a ?hacker home? that?s connected to Google?s Fiber broadband network. Synthia Payne uprooted from Denver and landed here to launch a start-up that aims to let musicians jam real-time online. That sleepy weekly gathering for Web entrepreneurs recently attracted a standing-room-only crowd of 260 businesspeople, investors and city officials.

Just as the move from dial-up modems to higher-speed Internet connections helped produce Netflix, Facebook and YouTube, policymakers and Google hope this next leap forward will breed a whole new slate of innovations.

The effort also is turning up the heat on cable companies, which now have to compete for consumers who can get faster speeds at lower monthly costs. Those telecom companies have begun bidding against Google to wire firms and city buildings with equally high-octane Internet.

?What Google is providing is a catalyst. This infrastructure is enormously important to create a ripple effect of entrepreneurial activity,? said Lesa Mitchell, a vice president at the Kauffman Foundation, a multibillion-dollar nonprofit that is trying to help local start-ups and officials turn around this city.

It?s an audacious and unproven experiment, the equivalent of replacing country roads with the Autobahn speedway and then assuming Formula One race cars will materialize. The question is whether it is a curiosity ? a publicity stunt ? or an example of what could happen around the country if more cities had access to such fast connections.

Some privacy advocates also worry that the project raises questions about how deeply Google will become entwined in its customers? lives.

?It gives them yet another way to gather and amass information about people, to build their digital dossiers,? said John Simpson, director at the public interest group Consumer Watchdog. ?They have so much data about users at their fingertips and become a magnet for government request for that information.?

But local officials think those lightning-fast Internet speeds, which allow movies to download in seconds and create picture- and sound-perfect video conference calls, will enable companies to operate more efficiently and use increased computing power to create cutting-edge technologies.

The ripples so far are small. About a dozen start-ups have been introduced in the first neighborhood to get Google?s 1-gigabit-per-second service.

Of course, Google has much to gain if the test in Kansas City works.

The first two calls we were back up plans for and we would have picked them up at the hospital. The two placements we have had, the CW dropped them off at the house. The CWs never checked out the bedrooms, but I am sure they checking to make sure the house is safe.

Oh, I didn't know, and it isn't a big deal for us, but at time of placement our agency does a quick background check on the foster parents. I'm sure it is just another safety net put in place to protect foster children.

Clay Guida has a high school wrestling meet to coach before fighting at UFC on Fox 6

CHICAGO -- Clay Guida will take on Hatsu Hioki on Saturday's UFC on Fox 6 card. It will mark Guida's first fight at featherweight, but before the bout and weigh-ins, he has another match on his mind.

Montini Catholic's wrestling team will face the Oak Park River Forest Huskies, and Guida will be matside to coach. He wrestled in high school and was part of a junior college national championship team at Harper College in suburban Chicago.

"I got a big wrestling meet tonight with the team I help coach, Montini Catholic at Oak Park-River Forest. We're no. 1, going for our sixth-straight state title. I believe they're no. 1 ranked in 3A, no. 2 in the state. Two top-10 teams in the nation."

Guida's longtime wrestling coach, Izzy Martinez, coaches at Montini, and he's brought fighters like Guida in to help his team. Guida's fight taking place in his hometown of Chicago days after Montini's biggest dual meet is lucky scheduling because it allows Guida to be at the meet. Working with the high school wrestlers gives Guida inspiration.

"It's a huge part of my training. These kids motivate me," Guida said during open workouts.

Guida says this fight is a new start for him. He was criticized by UFC president Dana White and fans for the loss to Gray Maynard for a safe fight strategy. Guida says that fight is behind him.

"You're going to see a more energetic, revamped Clay Guida. Educated fight fans see the strides I have made. I believe I'm one of the most improved lightweights, and soon to be featherweights, in the UFC. You see my striking. I outstruck Gray, I outgrappled Gray. I beat him everywhere except on the judges' scorecards. I'm beyond that now. It was a learning experience, and Hatsu Hioki is going to pay for it."

But before he gets in the cage with Hioki, it's the Montini Broncos' turn to perform, and Guida will be there coaching them on.

Modifications of a nanoparticle can change chemical interactions with cell membranes

Jan. 23, 2013 ? Researchers at Syracuse University's Department of Biomedical and Chemical Engineering at L.C. Smith College of Engineering and Computer Science are studying the toxicity of commonly used nanoparticles, particles up to one million times smaller than a millimeter that could potentially penetrate and damage cell membranes.

In a recent article published along with cover art in the journal Langmuir, researchers Shikha Nangia, assistant professor of biomedical and chemical engineering (BMCE), and Radhakrishna Sureshkumar, Department Chair of BMCE and professor of physics, showed how simple shape and charge modifications of a nanoparticle can cause tremendous changes in the chemical interactions between the nanoparticle and a cell membrane.

Nanomaterials, which are currently being used as drug carriers, also pose a legitimate concern, since no universal standards exist to educate and fully protect those who handle these materials. Nanoparticles are comparable to chemicals in their potential threat because they could easily penetrate the skin or be inhaled.

"Nanotechnology has immense potential that is starting to be being realized; a comprehensive understanding of toxicity of nanoparticles will help develop better safe handling procedures in nanomanufacturing and nano-biotechnology" says Sureshkumar and Nangia, In addition, the toxicity levels of various nanoparticles can be used to our advantage in targeting cancer cells and absorbing radiation during cancer therapy. Nanotoxicity is becoming a major concern as the use of nanoparticles in imaging, therapeutics, diagnostics, catalysis, sensing and energy harvesting continues to grow dramatically.

This research project has taken place over the past year utilizing a state of the art 448 core parallel computer nicknamed "Prophet" housed in Syracuse University's Green Data Center. The research was funded by the National Science Foundation.

Langmuir is a notable, interdisciplinary journal of American Chemical Society publishing articles in: colloids, interfaces, biological interfaces, nano-materials, electrochemistry and devices and applications.
